I am a Melburnian who went to school for four of my high school years in The Dandenongs.  Here are my favourite tips for a day trip from Melbourne to the Dandenong Ranges:

Sassafras is my favourite village.  Don’t miss: Miss Marples Tea Rooms, Gepetto’s Workshop, Up Art Gallery and Tea Leaves: The Crazy Teapot Shop.

Belgrave is my second favourite village.  I went to school here.  I used to think of it as a hippy town and it still is to some degree. 
The Cameo Cinema, as mentioned, shows great art house and mainstream films, the Puffing Billy is a major drawcard and nearby Sherbrooke Forest is a lovely way to have a picnic/bbq or hike in nature. 
Kerry Kulkens was on television back in the 70s and 80s giving her Numerology, Palmistry, Tarot and Clairvoyant predictions.  Her daughter was in my year level at school and now she is carrying on the witch tradition.  If you wish to have a private reading please book ahead.
The Sooki Lounge in Belgrave is a music venue that hosts a lineup of Australian and international acts.  It serves meals as well as craft beer, cocktails as well as great selection of gin all in a pretty chilled atmosphere.  Check out their gig guide

Bring good walking shoes because the walking and hiking tracks are beautiful and you never know what you might see – lyrebirds, cockatoos, lorikeets, wombats, echidnas and the like. 

In winter it can get very wet so make sure you bring wet weather gear.
